Both shots has taken with 100mm macro lens with same settings, the only difference is the number of images have used for stacking.
3 photos for the 1st picture and 10 photos for 2nd picture.
Settings :

Stacking on a moving object is almost impossible.
Congratulations on the second photo.
Ten shots!  And I can see that the fly was not dead.

Thanks Mandrake its my first try in extreme macro. planning to buy extension tube and ring flash to concentrate more in macro photography

ha ha, feeding fly is the solution, i used sugar syrups

Good shot. However, as Mandrake says , i too feel focus stacking is not the way to go for a moving object. you can get much more depth of field and sharpness , if your macro lens can stop down to f32, f48,etc and  you use ring flash to improve your shutter speed to 1/200 . 1/6 secs for a moving object is a bit too slow unless you want to show motion blur.

Completely agree Rama and Thank you for you comments, I have a plan to buy ring flash for macro photography
